Description: Epidermal cells from the inner surface of an onion flake. Beneath the shagreen-like cell walls one can see nuclei and small organelles floating in the cytoplasm. This BSE-image of a lanthanoid-stained sample was taken with EVO LS10 scanning electron microscope (Zeiss, Germany) without prior fixation, nor dehydration, nor sputtering. The unique method of sample preparation used here (BioREE, Russia, with support from the Research Institute of Eye Diseases, Russia) provides an outstanding contrast between subcellular structures giving an impression of light microscopy but at the resolution of SEM. A sample thus prepared resembles a block of ice with air bubbles in it – the deeper an object lies, the lower contrast it has and the more diffused it appears. Coauthor: A.M. Subbot
